OBJECTIVE: Angiotensin-(1-7) [Ang-(1-7)] is a pro-resolving mediator. It is not known whether the pro-resolving effects of Ang-(1-7) are sustained and protect the lung from a subsequent inflammatory challenge. This study sought to investigate the impact of treatment in face of a second allergic or lipopolysaccharide (LPS) challenge. METHODS: Mice, sensitized and challenged with ovalbumin (OVA), received a single Ang-(1-7) dose at the peak of eosinophilic inflammation, 24 h after the final OVA challenge. Subsequently, mice were euthanized at 48, 72, 96, and 120 h following the OVA challenge, and cellular infiltrate, inflammatory mediators, lung histopathology, and macrophage-mediated efferocytic activity were evaluated. The secondary inflammatory stimulus (OVA or LPS) was administered 120 h after the last OVA challenge, and subsequent inflammatory analyses were performed. RESULTS: Treatment with Ang-(1-7) resulted in elevated levels of IL-10, CD4+Foxp3+, Mres in the lungs and enhanced macrophage-mediated efferocytic capacity. Moreover, in allergic mice treated with Ang-(1-7) and then subjected to a secondary OVA challenge, inflammation was also reduced. Similarly, in mice exposed to LPS, Ang-(1-7) effectively prevented the lung inflammation. CONCLUSION: A single dose of Ang-(1-7) resolves lung inflammation and protect the lung from a subsequent inflammatory challenge highlighting its potential therapeutic for individuals with asthma.

During COVID-19 pandemic an increase electronic media (e-media) usage time was reported correlated to behavioral problems. This study verified the associations between e-media and behavioral/emotional problems in children in the context of the pandemic. Sample: Parents of 277 children aged 7-11 years old. Instruments: socioeconomic questionnaire; MAF-P and CBCL/6-18. Spearman's correlation analyses showed positive associations between the MAF-P and CBCL/6-18 scales. Adult games were correlated with 12 scales; websites for information/entertainment were associated with 6 scales, gambling online with one scale; electronic games and watching YouTube videos were associated with all 17 scales. In conclusion, overall, the results indicate the need of supervising children's e-media access considering the number and frequency of use and the elevated number of emotional and behavioral problems found among the evaluated group


Durante a pandemia da COVID-19 verificou-se aumento no tempo de uso de mídias eletrônicas (e-media) relacionado a problemas comportamentais. Este trabalho avaliou associações entre tempo de uso de e-media e problemas comportamentais/emocionais em crianças no contexto da pandemia. Contou com uma amostra de 277 responsáveis por crianças de 7-11 anos. Os instrumentos utilizados foram questionário socioeconômico, MAF-P e CBCL/6-18. As análises de correlação de Spearman apontaram associações positivas entre as e-media na MAF-P e escalas do CBCL/6-18. Os jogos adultos correlacionaram-se a 12 escalas; sites para informação/diversão foram associados a seis; apostar na internet a um; e jogos eletrônicos e ver vídeos no YouTube associaram-se a 17 escalas. Conclui-se, de modo geral, que os resultados indicam a necessidade de supervisão para acesso a e-media por crianças, considerando o número, a frequência de uso e o elevado número de problemas emocionais e comportamentais verificados no grupo estudado


Durante la pandemia de COVID-19, hubo un aumento en el tiempo dedicado al uso de medios electrónicos (e-media) relacionados con problemas de comportamiento. Este trabajo evaluó las asociaciones entre el tiempo de uso de los medios electrónicos y los problemas conductuales/emocionales en niños en el contexto de la pandemia. Muestra: 277 tutores de niños de 7 a 11 años. Instrumentos: socioeconómico, MAF-P y CBCL/6-18. Los análisis de correlación de Spearman mostraron asociaciones positivas entre los medios electrónicos en las escalas MAF-P y CBCL/6-18. Juegos para adultos correlacionados con 12 escalas; sitios de información/diversión asociados a 6; apostar en internet a las 1; los juegos electrónicos y ver videos en YouTube se asociaron con 17 escalas. Se concluye, en general, que los resultados indican la necesidad de supervisión para el acceso a e-media por parte de los niños, considerando el número y la frecuencia de uso y el elevado número de problemas emocionales y conductuales verificados en el grupo estudiado

This study investigated whether the origin of sperm (epididymal vs. ejaculate) affects the cryopreservation efficiency in agouti (Dasyprocta leporina). Five sexually mature agoutis underwent electroejaculation, resulting in obtaining four semen samples. After 15 days, the same animals were euthanized, and through retrograde flushing, sperm samples were obtained from the epididymis tails. In both collection methods, samples were evaluated for sperm parameters (sperm concentration, motility, vigor, membrane integrity, osmotic response, and morphology). Then, samples were diluted in ACP 109c, added with 20% egg yolk, and a final concentration of 6% glycerol. Finally, the samples were packaged in 0.25 mL straws and frozen in liquid nitrogen. After one week, samples were thawed and evaluated in the same way as fresh samples, with the addition of membrane integrity analysis using fluorescent probes (C-FDA/PI) and computerized analysis (CASA). Immediately after obtaining the sperm, samples obtained directly from the epididymis presented higher values (P ≤ 0.05) than those obtained by electroejaculation concerning the parameters of volume, sperm concentration, and total number of sperm (1,398.25 ± 206.0 x106 and 184.5 ± 78.0 x106 sperm). On the other hand, in the classical evaluation of the other sperm parameters and the computerized analysis (CASA) after thawing, such as total motility, no statistical differences were observed between sperm from both origins (ejaculate: 16.7 ± 8.2% and epididymal: 24.8 ± 12.0%, P > 0.05). This demonstrates the possibility of direct application of the cryopreservation protocol for agouti (D. leporina) sperm obtained via the epididymis or ejaculate.

Contradicting pediatric societies' recommendations, studies show that screen exposure begins at the first year of life for many children worldwide, with parental needs, educational purposes, and parental stress emerging as associated factors. However, the COVID-19 pandemic has likely worsened this scenario. This study aims to: 1) estimate the average daily screen exposure time for Brazilian infants aged 0-23 months during the COVID-19 pandemic based on caregiver report; 2) analyze the correlation between average exposure time, parental motivations for exposure, parental burnout levels and infant age; and 3) test the predictive role of parental motivations, burnout, and infant age on infant screen exposure. For this purpose, 139 parents (living in Brazil) answered online to questionnaires on infant screen exposure, parental motivations for exposure (four subscales: Parental Needs; Educational; Child's Desires/Family Routine; Behavioral Control), and parental burnout (Total score; four subscales: Emotional Exhaustion; Contrast; Feelings of Being Fed Up; Emotional Distancing). On average, infants were exposed to screens for 131 min/day. Higher scores on the four parental motivation subscales and higher parental burnout (specifically the Contrast subscale) correlated with longer exposure times. Furthermore, the motivation subscales Parental Needs and Child's Desires/family routine, the burnout subscale Contrast and infant age predicted greater screen exposure, while the burnout scale Feelings of Being Fed Up predicted less exposure. Our results suggest that infants in Brazil were exposed to substantial amounts of screen time during the pandemic. Interventions aiming to reduce screen time must consider parental motivations and parental burnout/mental health along with infant age.

The correlation between inattention and hyperactivity symptoms/behavior manifestations and screen time was evaluated among Brazilian children and adolescents (7-18 years old) who were socially isolated due to the COVID-19 pandemic. A total of 517 legal guardians completed questionnaires about electronic media use (MAF-P) and emotional/behavioral problems (CBCL/6-18). The results showed that texting was correlated to less inattention/hyperactivity symptoms; listening to music, the use of social media and electronics for school purposes were negatively correlated to attention problems and inattention/hyperactivity symptoms and playing videos games and online videos were associated to more attention problems and inattention/hyperactivity symptoms. The results contribute to the understanding of the relationship between inattention/hyperactivity symptoms and screen time in a sample of Brazilian children and adolescents during the pandemic.


Se evaluó la correlación entre síntomas/manifestaciones conductuales de inatención e hiperactividad y tiempo de uso de medios electrónicos entre niños y adolescentes brasileños (7-18 años) en aislamiento social por la pandemia de COVID-19. Los participantes fueron 517 tutores que completaron inventarios de uso de medios (MAF-P) y problemas emocionales/conductuales (CBCL/6-18), entre junio y agosto de 2020. Los resultados indican que más tiempo dedicado a la comunicación de mensajes se correlacionó con menos informes de síntomas de inatención/hiperactividad; Escuchar música durante más tiempo, usar redes sociales y dispositivos electrónicos para la escuela/el trabajo se asoció con menos síntomas inatención/hiperactividad y problemas de atención. Finalmente, jugar más videojuegos y ver videos en línea se asoció con más síntomas de falta de atención/hiperactividad y problemas de atención. Los resultados contribuyen a comprender que existen asociaciones entre la frecuencia de síntomas de inatención/hiperactividad y el tiempo de uso de medios electrónicos en una muestra de niños y adolescentes brasileños durante la pandemia.


Avaliou-se a correlação entre sintomas/manifestações comportamentais de desatenção e hiperatividade e tempo de uso de mídias eletrônicas entre crianças e adolescentes (7-18 anos) brasileiras em isolamento social devido a pandemia da COVID-19. Participaram 517 responsáveis que preencheram inventários de uso de mídias (MAF-P) e de problemas emocionais/comportamentais (CBCL/6-18), entre junho-agosto de 2020. Resultados indicam que mais tempo em comunicação por mensagens correlacionou-se a menos relatos de sintomas de desatenção/hiperatividade; já mais tempo ouvindo música, usando redes sociais e eletrônicos para escola/trabalho associaram-se com menor número de sintomas de desatenção/hiperatividade e de problemas atencionais. Por fim, maior em videogames e assistindo vídeos online associaram-se a mais sintomas de desatenção/hiperatividade e problemas atencionais. Os resultados contribuem para entender que existem associações entre frequência de sintomas de desatenção/hiperatividade e tempo de uso de mídias eletrônicas em uma amostra de crianças e adolescentes brasileiros durante a pandemia.

In pathological cardiac hypertrophy, the heart is more dependent on glucose than fatty acids. This shift in energy metabolism occurs due to several factors, including the oxygen deficit, which activates hypoxia-inducible factor-1α (HIF-1α), a critical molecule related to glucose metabolism. However, there are gaps regarding the behavior of key proteins in the glycolytic pathway and HIF-1α during the transition from hypertrophy to heart failure (HF). This study assesses the hypothesis that there is an early change and enhancement of HIF-1α and the glycolytic pathway, as well as an association between them during cardiac remodeling. Sham and aortic stenosis Wistar rats were analyzed at 2, 6, and 18 weeks and in HF (n = 10-18). Cardiac structure and function were investigated by echocardiogram. Myocardial glycolysis, the aerobic and anaerobic pathways and glycogen were analyzed by enzymatic assay, Western blot, and enzyme-linked immunosorbent assay (ELISA). The following were observed: increased left ventricular hypertrophy; early diastolic function change and severe systolic and diastolic dysfunction in HF; increased HIF-1α in the 2nd week and in HF; precocious alteration and intensification of glycolysis with a shift to anaerobic metabolism from the 6th week onwards; association between HIF-1α, glycolysis, and the anaerobic pathway. Our hypothesis was confirmed as there was an early change and intensification in glucose metabolism, alteration in HIF-1α, and an association between data during the progression from hypertrophy to heart failure.

OBJECTIVE: To translate the Fear Survey Schedule for Infants-Preschoolers (FSSIP) into Brazilian Portuguese and to examine its reliability and validity for assessing fears among Brazilian preschoolers. METHODS: Two independent bilingual professionals conducted translation and back-translation of the original survey. The translated version was used to assess 152 preschool children divided in two groups: Clinical - 71 children referred for treatment for nighttime fear, and Control - 81 children enrolled at kindergarten who had not been referred for any mental health service in the previous 6 months. All parents filled out the FSSIP, the Child Behavior Checklist (CBCL/1.5-5), and a sociodemographic questionnaire. RESULTS: Cronbach's alpha coefficients were 0.949 (95% CI: 0.94-0.96) for the entire sample; 0.948 (95% CI: 0.93-0.96) for the Clinical Group, and 0.95 (95% CI: 0.93-0.96) for the Control Group. The mean score for fears was higher in the clinical group (60.19 vs. 51.53, t = -2.056; p = 0.042), indicating acceptable discriminate validity. We also found positive, moderate, and statistically significant correlations between FSSIP and most CBCL scores, indicating good convergent validity. CONCLUSION: The Brazilian Portuguese version of the FSSIP showed good psychometric properties, and hence may be used in research and clinical settings to evaluate fears in preschoolers.

Introdução: O envelhecimento populacional cresce a cada ano e, consequentemente, surgem adaptações fisiológicas do organismo, tais como: diminuição da velocidade da marcha, da estabilidade e da adaptação postural, além da perda da integralidade muscular; podendo levar a várias alterações, dentre elas: disfunções do assoalho pélvico e prejuízo na mobilidade. Objetivo: associar a função do assoalho pélvico à mobilidade em idosas. Materiais e métodos: trata-se de um estudo transversal, desenvolvido com mulheres acima de 60 anos, residentes na cidade de Bauru/SP. O estudo foi aprovado pelo Comitê de Ética em Pesquisa (parecer nº4.040.883), e todas as voluntárias, que assinaram o Termo de Consentimento Livre e Esclarecido, responderam aos questionários para caracterização sociodemográfica e verificação das queixas de disfunções do assoalho pélvico. A avaliação da mobilidade foi realizada pela marcha, por meio de uma passarela de 14 metros e da análise dos parâmetros cinemáticos da marcha pelo módulo de aquisição de sinais biológicos, com um acelerômetro triaxial posicionado sobre o maléolo lateral e uma bateria de avaliação Short Physical Performance Battery (SPPB). Resultados: No total, foram avaliadas 13 mulheres (7 no grupo com disfunção do assoalho pélvico e 6 no grupo controle), com média de idade de 64 ± 6,3 anos. As disfunções do assoalho pélvico relatadas foram: 54% de incontinência urinária, 8% de prolapsos de órgãos pélvicos, 15% de incontinência fecal e 8% de disfunção sexual. Nos resultados do SPPB, 92% das voluntárias apresentaram boa capacidade; 8%, baixa capacidade. Na avaliação da marcha, não foram encontradas diferenças estatísticas entre os grupos (p=0,260). Conclusão: Não houve relação entre as funções do assoalho pélvico e a mobilidade nas participantes avaliadas.


Introduction: Population aging grows every year and, consequently, there are physiological adaptations of the organism such as a decrease in gait speed, stability, and postural adaptation, in addition to a loss of muscle integrality, which can lead to several changes, including pelvic floor dysfunctions and impaired mobility. Objective: to associate pelvic floor function with mobility in elderly women. Materials and methods: this cross-sectional study consisted of women over 60 from the city of Bauru /SP. The Research Ethics Committee (number 4.040.883) approved the study, and all volunteers signed the Informed Consent Form and responded to questionnaires for sociodemographic characterization and verification of complaints of pelvic floor dysfunction. Mobility assessment was performed by gait using the 14-meter walkway and analysis of the kinematic parameters using the biological signal acquisition module and a triaxial accelerometer positioned on the lateral malleolus and the evaluation battery Short Physical Performance Battery. Results: A total of 13 women were evaluated (7 in the group with pelvic dysfunction and 6 in the control group) with a mean age of 64± 6,3 years. 54% of women reported urinary incontinence, 8% prolapse, 15% fecal incontinence and 8% sexual dysfunction. In the results of the SPPB, 92% of the volunteers had good ability, and 8% had low ability. In the gait assessment, there were no statistical statistics between the groups (p = 0.260). Conclusion: There was no relation between pelvic floor function and mobility in the evaluated participants.

Studies on semen and sperm cells are critical to develop assisted reproductive technologies for the conservation of the collared peccary. The objective of the study was to compare the effect of different antibiotics on the bacterial load and sperm quality during short-term storage of peccary semen. Fresh semen samples from 10 males were extended in Tris-egg yolk or Tris-Aloe vera supplemented with streptomycin-penicillin (SP; 1 mg/mL - 1000 IU/mL or 2 mg/mL - 2000 IU/mL) or gentamicin (30 µg/mL or 70 µg/mL) before storage at 5°C. Bacterial load and sperm motility, membrane integrity and function, mitochondrial activity, and morphology, were evaluated at different time points for 36 h. The SP and gentamicin treatments concentration inhibited (p < 0.05) bacterial growth for 36 h regardless of the extender. Compared to the other treatments, Tris-egg yolk plus 70 µg/mL gentamicin maintained the sperm parameters for longer, including total motility (41.9 ± 6.1%) at 24 h, and membrane integrity (58.3 ± 2.1%) at 36 h. In contrast, the highest SP concentration in both extenders impaired sperm membrane integrity at 36 h (p < 0.05). For the liquid storage of collared peccary semen, it therefore is recommended to use Tris extender supplemented with egg yolk and gentamicin (70 µg/mL).

Insulin and insulin-like growth factors (IGFs) are mitogenic and prosurvival factors to many different cell types, including acute lymphoblastic leukemia (ALL). Circulating IGFs are bound by IGF binding proteins (IGFBPs) that regulate their action. IGFBP7 is an IGFBP-related protein (IGFBP-rP) that in contrast to other IGFBPs/IGFBP-rPs features higher affinity for insulin than IGFs and was shown to bind the IGF1 receptor (IGF1R) as well. The role of IGFBP7 in cancer is controversial: on some tumors, it functions as an oncogene, whereas in others, it functions as a tumor suppressor. In childhood ALL, higher IGFBP7 expression levels were associated with worse prognosis. Here we show that IGFBP7 exerts mitogenic and prosurvival autocrine effects on ALL cells that were dependent on insulin/IGF. IGFBP7 knockdown or antibody-mediated neutralization resulted in significant attenuation of ALL cell viability in vitro and leukemia progression in vivo. IGFBP7 was shown to prolong the surface retention of the IGF1R under insulin/IGF1 stimulation, resulting in sustained IGF1R, insulin receptor substrate 1 (IRS-1), protein kinase B (AKT), and extracellular signal-regulated kinase (ERK) phosphorylation. Conversely, the insulin receptor was readily internalized and dephosphorylated on insulin stimulation, despite IGFBP7 addition. The affinity of homodimeric IGF1R for insulin is reportedly >100 times lower than for IGF1. In the presence of IGFBP7, however, 25 ng/mL insulin resulted in IGF1R activation levels equivalent to that of 5 ng/mL IGF1. In conclusion, IGFBP7 plays an oncogenic role in ALL by promoting the perdurance of IGF1R at the cell surface, prolonging insulin/IGF stimulation. Preclinical data demonstrate that IGFBP7 is a valid target for antibody-based therapeutic interventions in ALL.

The aims were to identify the effects of growth differentiation factor 9 (GDF-9) on the in vitro development of ovarian preantral follicles (PAFs) of collared peccaries. Ovarian fragments were in vitro cultured for 1 or 7 days without or with inclusion of GDF-9 in the medium (0, 50, 100, or 200 ng/mL). The non-cultured (control) and cultured fragments were evaluated for PAF viability, activation, and cell proliferation. Although there were no differences in the percentage of morphologically normal follicles, the percentage of growing follicles was greater compared to the control in all treatment groups, especially those cultured with 200 ng/mL GDF-9 for 7 days (P < 0.05). The inclusion of GDF-9 in the medium did not interfere with PAF viability (P> 0.05); however, treatment with 200 ng/mL GDF-9 resulted in greater (P < 0.05) cell proliferation in PAFs cultured for 1 or 7 days (∼2.5 nucleolar organizing regions - NORs) compared to the follicles of the control group (2.0 NORs). In addition, peccary ovarian cortexes were subjected to PCR analysis and there was detection of the mRNA GDF-9 receptor transcripts of the BMPR2 (type I receptor) and ALK-5 (type II receptor) types. In conclusion, GDF-9, especially at a 200 ng/mL inclusion in the culture medium, was actively involved in the in vitro development of collared peccary PAFs.

Ovarian response of collared peccaries (Pecari tajacu), after hormonal stimulation with gonadotropin association (eCG/hCG), was accessed by both gene expression and follicular development. Thus, collared peccaries (n = 8) were treated with the dose used for sows (swine dose, SWD) or with dose adjusted for peccary's weight (allometric dose, ALD). The gene expression of receptors was evaluated for both gonadotropins (FSHR and LHCGR) and growth factors (proteins codified by TGFßR-1, BMPR1-A and BMPR2 genes) in antral follicles, cortex and corpora haemorrhagica (CH). Five days after gonadotropin injection, all females presented CH. The ovulation rate was similar (p > .05) between SWD (4.00 ± 1.17) and ALD (2.50 ± 0.43) group. The total number of follicles per animal and amounts of small (<3 mm), medium (3-5 mm) and large (>5 mm) follicles was similar among groups. However, SWD produced large follicles heavier than ALD group, as accessed by weight of follicular wall biopsies. Ovarian follicles expressed both gonadotropin and growth factor receptors at levels which are independent from gonadotropin dose. In conclusion, the two gonadotropin doses (SWD and ALD) can be used for ovarian stimulation of collared peccary. Additionally, FSH and growth factors (TGFßR-1, BMPR1-A and BMPR2) receptors are more expressed in the early follicle development, while LH receptor seems to be more important in the final of follicular growth.

This study investigated the effects of BMP-15 on the in vitro development of preantral follicles of collared peccaries. Ovarian fragments were cultured for 1 or 6 days in Tissue Culture Medium 199 (TCM199+ ) supplemented with BMP-15 at rates of 0, 1, 25 or 50 ng/ml. The fragments were analysed histologically by evaluating follicular morphology, activation and growth as well as the potential for proliferation of granulosa cells. Our results show the addition of 25 ng/ml BMP-15 in the medium provided the greatest percentage of normal follicles (79.67% ± 0.69) when compared to other treatments (p < .05); however, this result is similar to 1 ng/ml BMP-15 (74.00% ± 1.90, p > .05). Moreover, 25 and 50 ng/ml of BMP-15 promoted follicular activation. BMP-15 supplements did not affect oocyte and follicular growth. All concentrations of BMP-15 increased the number of nucleolus organizer regions (NORs) after 1 day of culture when compared to fresh fragments or the control samples (p < .05). However, at the end of the experiment, the number of NORs in follicles cultured in all treatments was higher than that observed in the fresh control (sample taken prior to culturing) (p > .05). In summary, the addition of 25 ng/ml BMP-15 to the culture medium of collared peccary preantral follicles maintained a high number of morphologically healthy follicles and stimulated the activation of primordial follicles after 6 days in culture.

In contributing to the conservation of wild rodents, the aim of this study was to evaluate the use of distinct cryoprotectants, separately or in combination, for solid surface vitrification (SSV) of red-rumped agouti ovarian tissue. Ovarian cortex from nine females was recovered and fragmented. Fresh fragments (control) were used to analyse the pre-antral follicle (PF) morphology using a histologic procedure, viability using the Trypan blue test, cell proliferation by counting the argyrophilic nucleolar organizing regions (Ag-NORs technique) and DNA integrity using the TUNEL assay. The remaining fragments were vitrified using SSV method with 3 M or 6 M ethylene glycol (EG) or dimethyl sulfoxide (DMSO), or in combination (3 M EG/3 M DMSO), and further evaluated as reported for the fresh samples. All cryoprotectants were effective at preserving PFs morphology compared to the control group (80.7 ± 5.21%), except 6 M EG and 3 M DMSO that provoked a significant (p < .05) decrease on the values of morphologically normal primary (60.0 ± 19.0%) and primordial (44 ± 4.5%) follicles, respectively. Regarding viability, all cryoprotectants provided values similar to that verified for the control group (79.0%), but a significant decrease (p < .05) was observed with EG/DMSO combination (59%). Using Ag-NORs technique, the highest (p < .05) cell proliferative capacity was detected when using EG at each tested concentration. The TUNEL proved the preservation of DNA integrity regardless of the cryoprotectant. In summary, we suggest the use of 3 M EG for the solid surface vitrification of red-rumped agouti ovarian tissue.

Due to the global decrease in jaguar population, conservation strategies are essential and the development of effective semen cryopreservation protocols would contribute to the formation of germplasm banks. Therefore, the objectives were to (1) evaluate the use of TRIS and ACP-117c extenders for jaguar semen freezing, (2) describe the ultrastructural changes in sperm after cryopreservation, and (3) evaluate the binding capacity of the thawed sperm. Eight ejaculates from five mature individuals were collected by electroejaculation, extended in TRIS or a coconut based-extender (ACP-117c), and frozen in liquid nitrogen. Samples were evaluated for sperm motility, vigor, membrane functionality, mitochondrial activity, morphology (using light microscopy, scanning electron microscopy - SEM and transmission electron microscopy - TEM), sperm kinetic parameters (by computerized analysis - CASA), and sperm binding capability using an egg yolk perivitelline membrane assay. Samples preserved in TRIS presented better post-thaw motility (46.0 ±â€¯7.7%) and membrane functionality (60.5 ±â€¯4.2%) and higher mitochondrial activity (21.5 ±â€¯3.7%) than those preserved in ACP-117c (20.9 ±â€¯5.4% motile sperm; 47.1 ±â€¯2.5% functional membrane; 11.8 ±â€¯1.7% mitochondrial activity). Regarding ultrastructural evaluations, SEM showed that both extenders were able to preserve the superficial membrane of the sperm, but TEM revealed the occurrence of nuclear electron lucent points, especially in samples extended in ACP-117c. Additionally, TRIS also provided a higher number of sperm bound to the perivitelline membrane (29.5 ±â€¯3.3%) in comparison to samples diluted in ACP-117c (18.6 ±â€¯1.5%). Overall, we suggest the use of a TRIS-based extender for cryopreservation of jaguar semen.

Interleukin 7 (IL-7) is a critical cytokine that plays a fundamental role in B- and T-cell development and in acute lymphoblastic leukemia (ALL). Its receptor (IL7R) is a transmembrane heterodimer formed by the IL7Rα and the IL2Rγ chain (γc). The IL7R signals through the JAK/STAT pathway. Loss-of-function mutations and some polymorphisms of the IL7Rα were associated to immunodeficiency and inflammatory diseases, respectively. Gain-of-function mutations were described in T-cell ALL and in high risk precursor B-cell ALL. Most confirmed loss-of-function mutations occur in the extracellular part of the IL7Rα while oncogenic mutations are exclusively found in the extracellular juxtamembrane (EJM) or transmembrane regions. Oncogenic mutations promote either IL7Rα/IL7Rα homodimerization and constitutive signaling, or increased affinity to γc or IL-7. This work presents a review on IL7Rα polymorphisms/mutations and attempts to present a classification based on their structural consequences and resulting biological activity.

The aim of this study was to evaluate environmental effects in a semiarid region on collared peccary seminal plasma content and sperm motility. Ejaculates from 12 mature males were obtained during the peak of rainy and dry periods of the Caatinga biome. Samples were evaluated for semen volume, pH, as well as sperm concentration, morphology, osmotic response, membrane integrity, chromatin condensation, and kinetic motility. Seminal plasma was evaluated for ions and organic compounds. The values for chloride, iron, magnesium, phosphorus, citric acid, cholesterol, triglycerides, total proteins, albumin, and fructosamine were similar during the dry and rainy periods; however, concentrations of fructose (849.2 mg/dL compared with 119.4 mg/dL) and calcium (32.3 mg/dL compared with 15.6 mg/dL) were greater during the rainy compared with dry period (P < 0.05). There were correlations (P < 0.05) among values for semen variables and biochemical contents, particularly between fructose and sperm velocity average pathway (r = 0.65), velocity straight line (r = 0.78), velocity curvilinear (r = 0.57), amplitude lateral head (r = 0.62), linearity (r = 0.41), and subpopulation with a medium velocity (r = -0.75). Furthermore, values for relative humidity were positively correlated with concentrations of fructose (r = 0.49), while air temperature (r = -0.43) and wind velocity values (r = 0.66) were negatively affected by concentration of fructose (P < 0.05). There were novel results regarding collared peccary seminal plasma biochemistry indicating there are important correlations with values for semen variables that are affected by the environment in a semiarid climate.

This study aimed to evaluate different vitrification methods using distinct cryoprotectants (CPAs) for the preservation of collared peccary ovarian preantral follicles (PFs). Ovarian pairs from six females were fragmented and three fragments (fresh control group) were immediately evaluated for morphology, viability, cell proliferation capacity (assessed by quantifying the number of argyrophilic nucleolus organizer regions - NORs), and apoptosis (by the identification of activated caspase-3 expression). The remaining 18 fragments were vitrified using the solid surface vitrification (SSV) method or the ovarian tissue cryosystem (OTC) with 3 M ethylene glycol (EG), 3 M dimethylsulfoxide (DMSO), or a combination of the two (1.5 M EG/1.5 M DMSO). After two weeks, samples were rewarmed and evaluated as described previously. The OTC with any of the CPAs provided a similar conservation of morphologically normal PFs as the fresh control group (75.6 ±â€¯8.6%); however, the SSV was only efficient with DMSO alone (63.9 ±â€¯7.6%). Regarding the viability or cell proliferation, all tested groups provided post rewarming values similar to those observed for the fresh control group, 84.0 ±â€¯2.9% viable cells with 2.0 ±â€¯0.2 NORs. Related to apoptosis analysis, only the OTC with EG (46.7%) and the SSV method with EG (43.4%) or the combination of EG and DMSO (33.4%) provided similar values to those found for the fresh control group (36.7%). Our findings indicate the utilization of a closed system, the OTC, with 3 M EG as the CPA for the vitrification of collared peccary ovarian tissue.
